Study on transfer of total soybean DNA into pigeonpea varieties

YANG Shiying, PANG Wen, LI Rongbai, LIANG Yuntao, HE Ziping, LU Weili, LUO Yuxi, CAI Qingsheng, CHEN Yanhua (Guangxi Academy of Agricultural Sciences, Nanning 530007,China)  
Total soybean DNA was transferred into pigeonpea varieties, ICPL 87091 and ICP 7035, through pollen tube channel. The D 3 transDNA plants showed variation in stem colour, flower colour, leaf colour and leaf type. Labeled with DIG High Primer DNA Labeling and Detection starter Kit II, the DNA of soybean variety Gui Pin 24-2 was used as probes to hybridize with DNAs from soybean variety Gui Pin 24-2 (donor), pigeonpea varieties ICPL 87091 and ICP 7035 (receptors) and their 12 selected transDNA progenies with better agrocharacters for whole genome DNA in situ hybridization analysis. Results showed strong DNA hybridization signal between some transDNA pigeonpea plants and soybean. Compared with the in situ hybridization performance of the donor and receptors, 7 transDNA pigeonpea progenies were clearly indicated high DNA homologous fragments, while the others showed low homologous fragments because the latter showed weak DNA hybridization signal with soybean probes. Our study results indicated some fragments of donor DNA have integrated into pigeonpea genome and expressed characters in the transDNA progenies.
