Effects of heat treating and some nutrient elements on the germination of basidspore of Volvariella valvacea

To learn the optimum condition for their germination, the basidspore of Volvariella valvacea were treated with different temperature and time of heat treatment, various mediums and carbon nutritive elements. The result showed that: the one treated with 45℃ for 30 min., the germination rate reached 8.7% while that of CK was only 1.2%, which indicated that proper heat stress may promote germination of the basidspore; PDA was better than CYM as their cultural medium for the germination; adding peptone and yeast extract into the media could accelerate the germination and the mycelium growth of the basidspore; the germination rate in the media with sucrose and maltose, as carbon nutrition, were obviously higher than that with lactose, and the later was significantly higher than that with glucose; the germinations rates of the 4 media were 32.1%, 30.0%,24.0% and 18.2%, respectively. Among the 4 media, the one with glucose was the best for the growth of the mycelium while the one with lactose was the worst.
