Preparation of Gramine under Ultrasonic Conditions

A ultrasound-assisted synthesis method of gramine was developed using dimethylamine,formaldehyde and indole as starting materials.Influences of catalysts,solvents,temperature,ultrasonic time,the mole ratio of starting materials to yields were investigated.When the reaction temperature is45℃,ultrasonic time is 5min,the catalyst is acetic acid,the molar ratio of dimethylamine,formaldehyde,indole and acetic acid was 2.0∶2.0∶1.0∶2.5,yield of gramine is 98.4%.The reaction was amplified to ten grams level.The products were characterized by 1 H NMR and IR.Compared with the existing methods,ultrasonic assisted synthesis of gramine has such advantages as easy operation,higher yield,shorter reaction time,which has a potential application prospect.
