HPLC method for determination of vitamin D_2 and vitamin E content in fatsoluble vitamin injection

Objective To develop the HPLC method for determination of vitamin D_2,vitamin E content in the Fat-soluble vitamin injection. Methods High performance liquid chromatography(HPLC) was used.Kromasil 100-5sil(250mm×4.6mm) chromatographic column was used to determine vitamin D2,n-hexane: isopropanol(97.5:2.5) was as mobile phase,the flow rate was 1ml/min,wavelength was 265 nm,under the condition of room temperature,and UV-detector were used.Kromasil 100-5sil(250mm×4.6mm) chromatographic column was used to determine vitamin E:n-hexane:isopropanol(99.5 ∶ 0.5) was as mobile phase,the flow rate was 1ml/min,the wavelength was 298 nm,under the condition of room temperature,and UV-detector were used. Results There was a good linear relationship between the 0.26-2.08μg/m L range and peak area for vitamin D2,while the 0.64-320μg/m L range for vitamin E.The average recovery rate of vitamin D2 and vitamin E were 98.90% and 100.2%.RSD of vitamin D2 and vitamin E were 1.2% and 1.1%. Conclusion The method is accurate,rapid and reproducible.
