Determination of Three Lignans in Schisandra sphenanthera Rehd. et Wils. from Different Producing Areas by LC-MS/MS

OBJECTIVE To establish an HPLC-MS/MS method for determination of schisandrin,deoxyschisandrin and schisandrin B contents in Schisandra sphenanthera Rehd. et Wils. from different producing areas. METHODS The samples were ultrasonically extracted with 95% ethanol for 10 minutes and separated on an Eclipse Plus C_(18) column( 2. 1 × 50 mm,3. 5μm) with gradient elution using a mobile phase that consisted of 10 m M ammonium acetate-methanol. The eletrospray ion-ization( ESI) with positive source of the mass spectrometer and detected in multiple reaction monitor( MRM) mode. RESULTS The three components of schisandrin,deoxyschisandrin and schisandrin B had favorable linearity in the ranges of 0. 61 ~ 5015 ng·mL~(-1),531. 9 ~ 6058. 8 ng·mL~(-1),0. 64 ~5230 ng·mL~(-1)( r≥0. 9944),respectively. This method showed an excellent recovery of 89. 3%,92. 5%,87. 8%with a RSD of 9. 85%,6. 40%,11. 23%,respectively. CONCLUSION This established method is simple sample preparation,high extraction efficiency and high sensitivity. The content of deoxyschisandrin in Schisandra sphenanthera Rehd. et Wils. is higher,the content of schisandrin and schisandrin B is lower,and the content of each area varies widely.
