Cloning and Analysis of the Evolutionary Tree's of Bovine Coronavirus HE Gene

The bovine coronavirus nucleic acid was extracted to study the bovine coronavirus HE gene evolution. The RNA was transcribed reversely into cDNA which was amplified as a template to a length of 1 300 bp HE gene fragment,connected with p MD-18 T vector,cloned into DH5α.It was sequenced and analysed the evolution tree with 14 strains BCV HE gene of the sequences published in Gen Bank. Amplification of bovine coronavirus HE gene sequence(HLJ-14),according with the characteristics of the HE genotype reference sequence,and Gen Bank had been published in the other 14 strains of BCV HE gene sequence homology of 97.3%-98.8%.
