Design of dsRNA Regions for Drosophila Cullin 5 Gene and Detection of the RNAi Efficiency

As one of the members of Cullin proteins family,Cullin 5 played an important role in regulation of protein degradation. To facilitate the further studies on the function of Cullin 5 in Drosophila,ds RNA targeted to the 5'UTR region of Cullin 5 was designed,and synthesized by PCR and the in vitro transcription methods. Then the RNAi efficiency was detected through RNAi and Real-time PCR. The results showed that the transcriptional level of Cullin 5 was reduced significantly by the ds RNA of Cullin 5 synthesized in this study(P0.01),and the m RNA level decreased to 27% compared to that in the control group. This study could provide strong technical support for the study on function of Cullin 5 in Drosophila in the future.
