Cloning of PtYbeY from Populus trichocarpa and Transformation into Arabidopsis thaliana

In order to explore the function of YbeY in populus,PtYbeY from populous was cloned by RT-PCR method,which was based on published poplar genome data. The gene was introduced into Arabidopsis by the floral dip method and homozygous transgenic plants expressing YbeY gene were identified. Sequence analysis showed that the CDS full-length of poplar PtYbeY gene was 1 761 bp and encoded 568 amino acids,which was contained UPF0054 domain and HAD hydrolase-like domain. The phenotype of transgenic plant by means of heterologous transformation was green of wild type plant compared to yellow of mutant plant. The phenotype analysis showed that PtYbeY could complemented the genetic function of in atybe Y mutant of Arabidopsis. The results of the study was indicated that the two homologous genes had the similar biological function,which contributed for further illustrating its function and molecular mechanism.
