Cloning and polymorphism analysis of Agouti gene exon 2 in reddish-brown Wusuli raccoon dog

In order to study cloing and polymorphism of Agouti gene exon 2 in reddish-brown Wusuli raccoon dog,3 wild-type and 3reddish-brown Wusuli raccoon dogs was selected as research object. Using cloning sequencing technique,the 156 bp DNA sequence of the exon 2 of Agouti was obtained to analyze the polymorphism and homology of the reddish-brown Wusuli Raccoon dogs,wild-type Wusuli raccoon dog and the other species. The results showed that the homologous rate of Agouti gene exon 2 in reddish-brown Wusuli raccoon dog and canis familiaris,felis catus,chimpanzee,sus scrofa,equus caballus and Ovis arieswas was 93. 94% 、83. 33% 、80. 47% 、78. 57% 、80. 36% 、77. 98% respectively. After comparing the sequence of Agouti gene exon 2 at 130 bp basic group in wild-type and reddish-brown Wusuli raccoon dog,we found there was a base substitution from G to T in the sequence of Agouti gene exon 2 in reddish-brown raccoon dog which leaded to the amino acid from tryptophan to glycine.
