Establishment of the method for detecting the immuno combarray of simian B virus antibody

The aim of the present study was to establish a rapid assay method for the antibody of simian B virus( BV). The prokaryotic expression product of BV32 gene was prepared by gene engineering technique. The recombinant BV32 protein was used as antigen diagnostic reagent.The immuno combarray( IC) method was established to specific detect the IgG antibody of BV in the simian serum. The results showed that the optimum coating amount of antigen was 0. 01 mg/m L. The prepared IC was all able to specifically detect the positive serum of BV. There was no cross reaction between the IC and serum of other viruses. 1 ∶400 times diluted BV positive serum could be detected sensitively by the IC. The coefficient of variation( CV) was less than 10% even if the reaction of the same was repeated for 3 times. 40 samples of monkey serum were detected by the immune detection method. The coincident rate was 98% with ELISA method,and the Kappa coefficient was 0. 997. It was indicated that shows that the IC method had a high sensitivity,strong specificity,good repeatability and good practicability.
