A preliminary study of relationship between ABCA1 and miRNA-758 in the porcine kidney cells( PK15 cells)

The aim of the present study was to examine the relationship of expressions between ATP binding cassette transporter A1( ABCA1)mRNA and miR-758 in PK15 cells. The PK15 cells were transfected with miR-758 mimic,miR-758 inhibitor and their negative controls,respectively. The relative expression levels of ABCA1 mRNA and miR-758 were measured by qRT-PCR post transfection 24 h. And then the relationship was explored. The results showed that the ABCA1 mRNA expression level was significantly increased in the PK15 cells transfected with the miR-758 mimic,miR-758 inhibitor and their negative controls( P0. 01). The miR-758 expression level of the PK15 cells transfected with miR-758 mimic was significantly higher than blank group( until 30 000 times,P0. 01). The other transfection groups showed lower miR-758 expression levels compared with the blank group,but there were no significant difference( P0. 05). The results indicated that the expression of ABCA1 mRNA in PK15 cells was increased by transfection of miR-758,miR-758 inhibitor and their respective negative control.The miR-758 mimic could promote the expression level of miR-758 in PK15 cells( until 30 000 times),but did not inhibit the expression of ABCA 1 mRNA in the cells. The expression of ABCA1 mRNA in PK15 cells was not affected or slightly affected by transfection of miR-758 mimic. The miR-758 inhibitor could did inhibit the expression of miR-758 and promote the expression level of ABCA1 mRNA in PK15 cells.
