Study on the determination on exact ovulation time of Kunming dogs

In order to accurately determine ovulation time of Kunming dogs for improving breeding efficiency of Kunming dogs at the optimal mating time,several methods were used in this experiment,including external observation,vaginal smear method,teasing,ovulation test kit,and hormonemeasuring for estrogen( E2),luteinizing hormone( LH),progesterone( P),also and ultrasonic diagnosis. Those methods were carried out to comprehensively find out the optimal ovulation time of Kunming dogs,and then surgical operation was conducted for the observation of ovary oviduct development and ovulationstatus,and experimentally verifying oocyte collection in vivo. The results showed that ovulation period of Kunming dog was during 48 ~ 72 h postthe peak of LH value,being 9 ~ 10 days post colporrhagia; keratinocyte cells invaginal smearreached 90%; Progesterone concentration was( 12. 79±1. 25) ng/m L,and the appearance of ovarian follicles during ultrasounddiagnosis. Accurate rate of oocyte collection was 80%,andoocyte maturationrate was 55. 0%; 4 oocytes in average per Kunming dog were acquired. The results indicated that ovulation time of Kunming dogs could be accuratelyascertaind according to the joint methods including estrus performance,vaginal smear,ultrasonic diagnosis,determination of LH and P. Oocyte collection in vivo was carried out by surgery,and matured oocytes could be collectedfrom theoviduct.
