The maize pollens were used to extract total RNA using 3 different methods(Trizol, acid guanidinium isothiocyanate-phenol-chloroform and modified CTAB) and their extraction effects were compared, with reference to the RNA isolation method for other plants. The agarose gel electrophoresis, instead of formaldehyde denaturalization electrophoresis, was used to analyze the integrality of RNA extracted by 3 methods, and the UV-spectrophotometer was used to analyze the purity of RNA.The result showed that the modified CTAB method was the best.
