Minitab Application in Studies on Medium Optimization of Avermectins Fermentation

Optimization of culture medium for avermectins fermentation with a Streptomyces avermitilis strain producing high avermectin B components was carried out by the'response surface method'(RSM) of Minitab software.The impact of initial medium constituents was studied by Plackett-Burman,which revealed that corn starch,soybean meal and CoCl2·6H2O have substantial influence on avermectin B1a components.Optimum concentrations of corn starch,soybean meal and CoCl2·6H2O were confirmed via Box-Behnken.The optimal medium consisted of(g/L): corn starch 92.19,peanut meal 10,soybean meal 9.70,yeast powder 5,yeast extract 2.8,corn syrup 2.2,CoCl2·6H2O 0.04.Under the optimal medium,the yield of avermectin B1a components reached 525μg/mL,an increase of 45% more than the original medium.
