In Situ Hybridization Analysis of Two Rice OsRhoGDIs Genes in Young Panicle

Rice OsRhoGDI1 and OsRhoGDI2 genes were newly isolated from young panicle by yeast two-hybrid system,which encoded the OsRacD-interacting proteins.In order to study the expression characteristics of OsRhoGDIs genes within rice young panicle,two probe templates were prepared by PCR amplification,and labeled by digoxigenin tagging using the random primer labeling method.Then the two probes were utilized to hybridize with paraffin sections of rice young panicle tissue.The result revealed that the transcript level of OsRhoGDI2 gene were obviously higher than that of OsRhoGDI1 gene,indicating that OsRhoGDI2 was probably the main protein regulating OsRacD activity.
