Bioinformatics Analysis of Pivotal HMGR Gene Involved in Essential Oil Biosynthesis of Dysphania schraderiana

The physical and chemical properties,secondary structures,signal peptide,transmembrane domain,subcellular localization,conserved domain,tertiary structure and evolutionary relationship of Ds HMGR protein were predicted and analyzed by a series of bioinformatics software. As results,Ds HMGR gene contained an open reading box of 1 725 bp. Ds HMGR was composed of 574 amino acids,among which most were non-polar amino acids. Ds HMGR was an unstable protein with the p I of 7. 46. In addition,Ds HMGR possessed two transmembrance domains anchoring the deduced protein to endoplasmic reticulum and two hydrophobic areas,but no signal peptide was found. The main secondary structure of Ds HMGR was α-helix spread throughout the peptide chain. The conservation in the C-terminus region with four conserved active sites and high variation in the N-terminus region compared with other plants were observed and might provide similar catalytic activity and disparate regulatory process of HMGR activity. Tertiary structure prediction resulted in the observation of three catalytic domains. Ds HMGR was clustered into Chenopodiaceae plants,indicating the closest phylogenetic relationship.
