Identification and Screening of Optimal Strain and Formula of Wild Termitomyces

In order to protect and utilize the wild germplasmic resources of Termitomyces,based on wide collection of wild test strains of Termitomyces from Lincang,the internal transcribed spacers( ITS) were sequenced,and analyzed systematically combined with the existing Gen Bank sequences to establish the taxonomy of them,furthermore,the optimal strain and formula were also preliminarily studied. The results showed that the test strains clustered together with other species of Termitomyces in the phylogenetic tree which were divided into two major branches and four minor branches. Test strains showed different growth characteristics on YPD medium,among which strain 4279 showed the best,followed by strain 2871 and strain 2878,and strain 4288 was the worst. Strain 4279 could grow on 11 test media. On the medium of cow dung,strain 4279 grew densely and had a strong growth potential,up to( 0. 77 + 0. 04) mm/d,which was significantly higher than the other media. In conclusion,ITS sequence analysis could distinguish the mixed Termitomyces,and be used as a method for classification and identification of macrofungi. The optimal formula for the mycelia growth of strain 4279 was 150 g cow dung + 20 g glucose + 15 g agar +2 g peptone,and the medium components had significant effect on the mycelial growth.
