Effect of Xintongtai on serum levels of endothelin-1 and nitric oxide in a rat model of myocardial infarction

Objective: To investigate the effect of Xintongtai on the serum levels of endothelin- 1( ET- 1) and nitric oxide( NO) in a rat model of myocardial infarction. Methods: The rat model of myocardial infarction was established,and the rats were divided into model group,sham- operation group,low-,middle-,and high- dose Xintongtai groups,and Shexiang Baoxin Pill group. The rats in the model group and the sham- operation group were given the same volume of distilled water by gavage,and the rats in the other groups were given respective doses of drugs by gavage. The serum levels of ET- 1 and NO and the area of myocardial infarction in rats were measured after 2 weeks of administration. Results:Each administration group had a significant reduction in the area of myocardial infarction compared with the model group( P 0. 01); compared with the low- dose Xintongtai group,the middle- and high- dose Xintongtai groups and the Shexiang Baoxin Pill group had significant reductions in the area of myocardial infarction( P 0. 01),while the area of myocardial infarction showed no significant differences between the three groups( P 0. 05). Compared with the sham-operation group,each treatment group and the model group had significantly increased ET- 1 expression and significantly decreased NO expression( P 0. 01); compared with the model group,each treatment group had significantly decreased ET- 1 expression and significantly increased NO expression( P 0. 01); compared with the low- dose Xintongtai group,the middle-,and high- dose Xintongtai groups and the Shexiang Baoxin Pill group had a significantly better improvement in the expression of ET- 1 and NO( P 0. 01),while there were no significant differences in this aspect between the three groups( P 0. 05). Conclusion: Xintongtai Granule can regulate the expression of ET- 1 and NO to inhibit vascular remodeling,increase coronary blood flow,and reduce the area of myocardial infarction in rats.
