Influence of pretreatment with Rougan Tongluo decoction on brain cell apoptosis and neuron morphology in rats with middle cerebral artery occlusion

Objective: To investigate the influence of Rougan Tongluo decoction on brain cell apoptosis and neuron morphology in rats with cerebral ischemia / reperfusion injury induced by middle cerebral artery occlusion( MCAO). Methods: The experimental rats were divided into high-,medium-,and low- dose Rougan Tongluo decoction groups,blank control group,sham- operation group,and model group. The suture method was used to establish the rat model of cerebral ischemia / reperfusion injury,and the terminal deoxynucleotidyl transferase- mediated d UTP nick- end labeling( TUNEL) method and Nissl staining were used to observe brain cell apoptosis and neuron morphology at 3,6,12,24,and 48 hours after ischemia / reperfusion. Results: Rougan Tongluo decoction significantly reduced the number of apoptotic cells and increased the number of viable neurons in rats. Compared with the model group,the Rougan Tongluo decoction groups showed a significantly lower number of apoptotic cells and a significantly higher number of viable neurons( P 0. 05). Conclusion: Rougan Tongluo decoction exerts its protective effect on the brain through fighting against cell apoptosis.
