Objective: To investigate the therapeutic effect of saikosaponin- d in guinea pigs with cough variant asthma( CVA) and related mechanisms of action. Methods: A total of 32 guinea pigs were randomly divided into normal group, model group,saikosaponin- d group, and hormone group. The comprehensive method was used to establish the guinea pig model of CVA and the corresponding drug intervention was given. The signs, times of cough, and eosinophil count in bronchoalveolar lavage fluid were observed in all guinea pigs, and the changes in the serum levels of interleukin- 5( IL- 5) and tumor necrosis factor- α( TNF- α) were determined. Results: Compared with the normal group, the model group experienced paroxysmal cough, sneeze, tachypnea, and restlessness / uneasiness and showed significant increases in the times of cough( P 0. 01),eosinophil count( P 0. 01),and serum levels of IL- 5 and TNF- α( P 0. 05). After the corresponding drug intervention was given, the saikosaponin- d group and the hormone group showed significant reductions in the times of cough, eosinophil count, and serum levels of IL- 5 and TNF- α( P 0. 05,P 0. 01). Conclusion: Saikosaponin- d can reduce the expression of IL- 5 and TNF- α in guinea pigs and thus achieve the goal of treating CVA.
