Effect of Shuangdan Mingmu capsules on the mRNA expression of Akt and endothelial nitric oxide synthase in rats with diabetic retinopathy

Objective: To investigate the effect of Shuangdan Mingmu capsules on the mRNA expression of Akt and endothelial nitric oxide synthase(eNOS) in a rat model of diabetic retinopathy. Methods: A total of 33 GK rats were used to establish a model of diabetic retinopathy and then they were randomly divided into positive drug control group,Shuangdan Mingmu capsule group(12. 5 ml/kg),and model group,with 11 rats in each group. Another 11 healthy Wistar rats were included in blank group. The rats were treated with the drug by gavage since one week after the model was established for2 months. RT-PCR was performed to measure the mRNA expression of Akt and eNOS. Results: Compared with the blank group,the model group had significantly higher mRNA expression of Akt and eNOS(P 0. 05). Compared with the model group,the positive drug control group and the Shuangdan Mingmu capsule group had significant reductions in the mRNA expression of Akt and eNOS(P 0. 05),and there were significant differences between these two groups and the blank group(P 0. 05). Compared with the positive drug control group,the Shuangdan Mingmu capsule group had significantly milder reductions in these indices(P 0. 05). Conclusion: Shuangdan Mingmu capsules treat and prevent diabetic retinopathy possibly by inhibiting the mRNA expression of Akt and eNOS and preventing pathological neovascularization.
