Isolation of Brucella on Pet Dogs as well as Its 16S rDNA Sequence Identification

A strain of bacteria from the whole blood of a suspected Brucella-infected pet dog was isolated,purified, stained and microscopically examined, and the 16 SrDNA gene fragment was amplified by PCR. The amplified product was purified and sequenced, and then NCBI software was used. The results showed that its homology with Brucella reached 100%; 16 SrDNA sequence similarity analysis was performed by using DNAStar software, and it was found that the nucleotide sequence similarity between the isolated strain and Brucella 16 S rDNA was between 99.6% and 100%, indicating that the isolated strain was Brucella, but whether the isolated strain was B. canis or B. bovis remains to be further studied.
