Effects of rhOGP on the decrease of white blood cell in cyclophosphamide-induced leucopenic mice

Objective To study the effects of rhOGP on the decrease of white blood cell in cyclophosphamide-induced leucopenic mice;To investigate the effects of rhOGP on the proliferation of human pulmonary adenocarcinoma cell line Anip and human gastric carcinoma cell line SGC-7901.Methods The leucopenic mice model was induced by an intraperitoneal injection of cyclophosphamide(Cy) for 3 consecutive days.In the antagonistic part,including the first 3 days of Cy injection,rhOGP(high and low doses)were given to the leucopenic mice for 13 consecutive days by subcutaneous administration.WBC were counted before administration at day 1,9,and 14,respectively.In the apotropaic part,doses of rhOGP,DTT,and 0.9% NaCl(aq) were given to the mice for 10 consecutive days.The leucopenic mice models were induced by an intraperitoneal injection of Cy for the last 3 days(day 8,9,and 10).WBC were counted before and after administration.The effects of rhOGP on the proliferation of Anip and SGC-7901 cells were determined by MTT assay and cell growth curve.Results The peripheral white blood cell count of low-dose-rhOGP group was significantly higher than that of the normal control group,and the apotropaic effects were visible.rhOGP could not promote the proliferation of Anip and SGC-7901 cells.Conclusion rhOGP could significantly enhance the restoration of WBC on Cy-induced aplastic mice.rhOGP has visible apotropaic effects on decreasing of WBC on Cy-induced aplastic mice.rhOGP could not promote the proliferation of human Anip pulmonary adenocarcinoma cell and human SGC-7901 gastric carcinoma cell.
