The mechanism of TFPI gene transfer mediated by adenovirus on the apoptosis of vascular smooth muscle cells

Objective To investigate the effect of tissue factor pathway inhibitor(TFPI) gene transfer on the pathway of apoptosis in vascular smooth muscle cell(VSMC) and approach the mechanism of its inhibition on cell apoptosis.Methods Human TFPI or LacZ recombinant adenovirus or DMEM was used to infect rat aortic VSMC respectively in vitro.ELISA was used to detect the expression of exogenous TFPI gene.Western blot was used to examine JAK-2,p-JAK-2,STAT-3,p-STAT-3 and cyclinD1 at different time points after gene transfer.Results Expression of TFPI protein was detected in VSMC at 1st day after gene transfer and the peak expression was at the 3rd day.The expressions of JAK-2 and STAT-3 had no significant differences in each group at the 3 rd,5th and 7th day after gene transfer(P0.05),but the expressions of p-JAK-2,p-STAT-3 and cyclinD1 in TFPI group were reduced compared with those in LacZ and DMEM groups(P0.05) in time dependent manner.Conclusion TFPI could induce apoptosis in VSMC through the inhibition of JAK-2/STAT-3 pathway,and therefore prevent occurrence of restenosis.
