Amines and peptides are important substances of biological activity in crustaceans. In the life process they play the role of hormone, neurotransmitter and/or neuromodulator, the optical ganglia and the brain are the center of neuroendocrinology in crustaceans, exerting significant modulation in molting, reproduction and other physical activities. Penaeus vannamei, despite being of global commercial importance, has never been subject to neuroendocrinological research. In this paper, immunocytochemistry techniques were used to observe the immunoreactive neurons and neuropils in the optic ganglia and brain of P . vannamei. The sections were stained with antibodies of serotonin (5 HT), neuropeptide Y (NPY), β Endorphin(β Ep), substance P (SP), somatostatin (SS), glucagon (Glu) and proctolin (PRL). The immunostained sections were observed and the positive neurons and neuropils were examined under a light microscope. Immunoreativities are found against 5 HT, NPY, β Ep and SP. Their distributions are as following: 5 HT immunoreactive cells are located in lamina ganglionaris (LG), medulla externa (ME), medulla interna (MI) and medulla terminalis (MT). The immunoreactivity of 5 HT was detected in the anterior median cluster, the anterior medial protocerebrum neuropiles, the inner and outer lateral cluster to olfactory lobe of the deutocerebrum. The presence of NPY immunoreactive cells was found in the four neuropils of the optic ganglia. The immunoreactivity of NPY was located in the anterior median cluster and the anterior medial protocerebrum neuropiles. SP immunoreactive cells and β Ep immunoreactive cells existed in the anterior median cluster of protocerebrum and the posterior lateral cluster of tritocerebrum respectively. The wide distribution of 5 HT immunoreactivity in the optical ganglion of P. vannamei suggests that 5 HT probably plays the role of neuromodulator in the nerve cells. The 5 HT immunoreactivity in the cerebrum of P. vannamei may supply morphological proofs for its engagement in the secretion of GSH (gonad stimulating hormone). The specific distribution patterns of immunoreactive substances against NPY, β Ep, SS and SP may also supply morphological proofs for their different neurophysiological functions. No immunoreativities were found against SS, Glu and PRL. However, it cannot be concluded that these 4 corresponding antigen do not actually exist in the nervous system of P. vannamei . Further investigation is needed to reduce the technical and biological limits of this research.
