Effects of AduoLa Fuzhenglin on changes of cytochrome c oxidase in rats′hippocampus after microwave exposure

Objective To investigate the effect of a herb compound AduoLa Fuzhenglin(ADL) on the gene cytochrome c oxidase(COX) change of the mitochondrial respiratory chain in rats′ hippocampus after microwave exposure.Methods One hundred Wistar male rats were exposed to microwave at the average power density of 30 mW/cm2for 15 min,3 g/(kg·d) ADL was administrated for 14 days before or after radiation.Rats were sacrificed at 6 h and 14 d after exposure.The activity of COX was determined by colorimetry;real time polymerase chain reaction was used to study the expression of COX Ⅰ and Ⅳ mRNA;the COX Ⅰ protein was detected by Western blot and image analysis.Results The level of COX activity,COX Ⅰ and Ⅳ mRNA and COX protein was decreased obviously in the radiated control group(P0.05 or P0.01) at 6 h and in the prophylaxis and treatment groups after drug withdrawal.The activity and gene expression of COX in the ADL prophylaxis group were higher than those in the radiated control group,and were not significant different from the normal control.Compared with the radiated control group,those in the ADL treatment group were significantly increased(P0.05),and almost returned to normal.Conclusion Administration of 3 g/(kg·d) ADL could lessen the decrease in the COX expression level of the mitochondrial respiratory chain in rats′ hippocampus after microwave exposure,and with better effect on ADL treatment group.
