STUDIES ON THE KINETIC PROPERTY OF ACID PROTEINASE FROM ASPERGILLUS USAMII MUTANT L336

The L336 acid proteinase from Aspergillus usamii was shown to have a molecular weight of 54000 and an isoelectric point of 3.8± 0.2. The Km Values of the enzyme at 40℃ and pH2.5 for casein、bovine serum albumin and bovin haemoglobine were 0.263g / L. 0.278g / L and 0.415g / L reapectively, and the Vmax values under the same conditions were 2075. 1550 and 2793μg Tyr / min、mL. The optimum substrate for acid proteinase was casein. The enzyme was stable at temperature up to 40℃ and pH2.5~5.0. In addition, the effect of different metal ions on the activities of enzyme was also studied.
