Isolation, Purification and Kinetics of CD26 from Porcine Kidney

Objective:To purify CD26 from porcine kidney and to study some of its properties.Methods:Affinity chromatography based on Sepharose 4B immobilized adenosine deaminase was used and enzyme activity was assayed according to the method of Egawa.The results showed that it was purified 341-fold with a 31.3% yield to homogeneity.The molecular weight of the purified CD26 was approximately 109.2 KD by SDS-PAGE.Furthermore,we obtained the kinetic parameters Km=(101.5±2.2) μmol/L and Vmax=(19.5±0.3) U/mg protein by using nonlinear regression analysis.Conclusion:CD26 can be purified to homogeneity by ADA-Sepharose 4B affinity chromatography.
