The Effect of Zinc on the caspase-9 Expression in Capsaicin Induced Pain Model Mice Spinal Cord

Objective To observe the effect of zinc on the expression of caspase-9 protein in capsaicin induced pain model mice spinal cord.Methods The C57/BL6 mice were pretreated with high level or low level of zinc ions for 2 weeks firstly.Then capsaicin was injected into the foot sole to build neuropathic pain(NPP) mice model.Immunohistochemistry,immunoblotting and image analysis were used to test the effect of zinc on the caspase-9 expression in NPP model mice spinal cord.Results The expression of caspase-9 protein and its optical density increased in spinal cord in every NPP model mice group(P0.01).The caspase-9 protein expression and its optical density decreased in high-zinc pretreated mice spinal cord,while increased in low-zinc pretreated mice spinal cord(P0.01).Conclusion Zinc ions could inhibit the caspase-9 protein expression in NPP model mice spinal cord and the zinc ions might take part in the development and conduction of NPP.
