Influence of Candesartan Cilexetil on the Oxidative Stress of Kidney and MCP-1 Expression in Diabetic Rats

Objective To observe the effects of candesartan cilexetil on oxidative stress of kidney and expression of monocyte chemoattractant protein(MCP)-1 in diabetic rats.Methods Ten out of forty healthy male SD rats were randomly selected as the control group(A).The other thirty rats given streptozotocin(STZ) were made as the model of diabetic rats,and they were divided into 3 groups with 10 rats in each: diabetic model group(B),the low doses of candesartan group(C) 5 mg /(kg.d),and high doses of candesartan group(D) 10 mg /(kg.d).After 12 weeks,the blood sugar,urea nitrogen level,kidney quality index,serum,superoxide dismutase(SOD) activity of renal tissue and content of malonaldehyde(MDA) were measured.The expressions of MCP-1 in renal tissue were investigated by immunohistochemistry and Western Blot.Results Compared with the control group,blood sugar and serum urea levels in the diabetes mellitus(DM) group and candesartan group increased(P 0.01),kidney quality index increased(P 0.01),content of MDA increased,SOD activity decreased(P 0.05),MCP-1 expression in kidney tissue increased(P 0.01).Compared with DM group,serum urea levels decreased(P 0.01),kidney quality index decreased(P 0.01),content of MDA decreased,SOD activity increased(P 0.05),MCP-1 expression in kidney tissue decreased(P 0.05) in the candesartan groups.Conclusion Candesartan has protective effect on diabetic rats’kidney injury induced by STZ.The protective effect might be related to the inhibition of oxidative stress and the reduction of MCP-1 expression.
