Calcitonin gene-related peptide regulates rat osteoclasts formation in vitro

Objective:To investigate the effect of calcitonin gene-related peptide on the formation of rat osteoclasts in vitro.Method:Rat bone marrow cultures were stimulated to generate osteoclasts using 1,25-dihydroxyvitamin D3; Effect of calcitonin gene-related peptide on the formation of rat osteoclasts were detected by observing the number of TRAP-positive multinucleated and mono-binucleated cells.Result:Calcitonin gene-related peptide dose-dependently decreased the numbers of TRAP-positive multinucleated cells as well as TRAP-positive mono-/binucleated cells at concentrations 10 -9 mmol/L、10 -8 mmol/L、10 -7 mmol/L(P0.05). Conclusion:The findings suggested that calcitonin gene-related peptide may inhibit osteoclasts development in bone marrow cultures.
