Establishment of in vitro culture of condylar cartilages

Objective:The purpose of this study was to establish the organ culture system and to investigate the biological characteristics of mandibular condylar cartilages cultured in vitro.Method:12 neonatal rats were selected and killed by shearing their heads.Both mandibular condyles with part of the ramus were removed aseptically from the tempotomandibular joints.The MCC was dissected,fragments of attached muscles and underlying bone were carefully removed.Then the MCC isolated was cultured in a modified Trowell-type system.The explants were placed on a membrane filter at the medium-gas interface and cultured for 6 days.Every day four cartilages were fixed and embedded in paraffin,sections strained by eosin and haematine.Result:Light microscopic observations showed that the mandibular condylar cartilages remained normal during organ culture.Within 4 days,the cells of the cartilages were integral.With culture time lasting,small cavities appear among the cells on the surface of the condylar cartilage.Then local cells on the surface layer began to degenerate.Conclusion:It is concluded that the primary MCC of SD rats cultured in vitro can constantly remain the normal structure of condylar cartilage,and the cells of these cartilages act biologically,so the cartilages cultured within this period can be used as the subjects to study the physiology and pathology of MCC in vitro.
