Research about biological properties of PDLSCs during physiological root resorption in deciduous tooth

Objective:To compare the biological properties of PDLSCs from deciduous tooth with different stages of physiological root resorption and to detect their expression of key factors related to the formation and apoptosis of osteoclast.Method:PDLSCs were isolated and cultured from deciduous tooth with different degrees of root resorption and permanent tooth. The study focused on their immune phenotype,proliferation and mutidifferentiation properties by flow cytometry,MTT assay and osteogenic/adipogenic induction respectively. Further as assayed by Western blot,the study examined the expression of RANKL,OPG and FasL,which are key factors to osteoclast status,of PDLSCs from each group. Result:PDLSCs from all these groups expressed mesenchymal stem cell surface markers and had strong proliferation ability. PDLSCs from the severely resorpted tooth had the strongest osteogenic differentiation and the weakest adipogenesis differentiation potential.The expression level of RANKL,OPG and FasL protein of these PDLSCs were different. Conlusion:Biological properties of PDLSCs from different stages of physiological root resorption were different. The different expression pattern of RANKL,OPG and FasL may contribute to the formation and apoptosis of osteoclast in the proscess of physiological root resorption in deciduous tooth.
