The effects of scutellarin on the expression of OPG mRNA and RANKL mRNA in rat osteoblast in vitro

Objective: To study the effection of scutellarin with different concentrations and different intervention time on the expression of OPG mRNA and RANKL mRNA in osteoblasts in vitro,thus to propose the role that scutellarin can play in bone rebuilding. Methods: Twenty new born wistar mice were taken their calvaria bone by asepsis and primarily cultured osteoblasts by trypase-collagenase enzyme digestion method. The third passage osteoblasts were divided into the control group and the different experiment group. The different scutellarin concentrations including 1 /10,1 /100,1 /1000 and 1 /10000 dilution concentration were respectively into the different experiment group. Total RNA was respectively isolated from osteoblasts after treatment with different concentration of scutellarin for 24,48 and 96 hours. Expressions of OPG mRNA and RANKL mRNA were detected by semiquantitative RT-PCR,and the value of OPG / GAPDH,RANKL / GAPDH were contrasted among the different groups. Results: The expression of OPG mRNA,RANKL mRNA and the value of OPG / GAPDH,RANKL /GAPDH in the different groups were changed in a time-and-dose dependent fashion. With the increasing of time and concentration,the expression of OPG mRNA and the value of OPG / GAPDH were increased gradually,while the expression of RANKL mRNA and the value of RANKL / GAPDH were decreased gradually. Conclusion: The results suggest that scutellarin could promote bone formation in some dug concentrations and some intervention time.
