Comparison of induction of model of cleft palate in mice between high and low doses of dioxin combined with dexamethasone

Objective: To explore the differences of induction of model of cleft palate in mice between different doses of dioxin combined with dexamethasone. Methods: C57 BL/6 J experimental mice were randomly divided into two groups,one group treated with continuous administration of TCDD( 3 μg/kg) + DEX( 6 mg/kg) for three days,and another group treated with high doses of TCDD( 6 μg/kg) + DEX( 25 mg/kg) for one time. The incidence of cleft palate,the rate of abortion and the rate of stillbirth were recorded and analyzed statistically. Results: The incidence of cleft palate in mice was 100% in both of the two groups,and the rate of stillbirth in high dose group was lower than that in low dose group,and the difference was statistically significant. Conclusion: The effect of teratogenicity of high dose TCDD on pregnant rats is less,the rate of fetal stillbirth is significantly reduced,the abortion rate of pregnant mice is low,the model of cleft palate is more stable,and it is more suitable for follow-up study.
