Change of 11β-HSD1 Expression in Visceral Bed Fat of Diabetic Rate

Objective: To assay the effect of 11β-hydroxysteroid dehydrogenase type 1 of visceral bed fat on diabetic rats. Methods: The experimental rats were divided into five group: normal rats (NC), fat-diet-induced rats (FD), fat- diet- and STZ-induced diabetic rats (DM), rosiglitazone-treated DM rats (RDM) and carbenoxolone-treated DM rats (CDM). The expression of 11β-HSD1 mRNA and protein were observed in each group respectively. After performed IPGTT, serial plasma glucose, insulin were detected. TG, TCh, Fast plasma cortisone were also detected. Results: The expression of 11β-HSD1 in visceral bed fat was higher in DM, RDM, CDM and FD than in NC,and was correlated with fast plasma F, insulin, TG, TCh and AUC-I/G;Concentrations of Fasting plasma F, TG and insulin were lower in treatment of CDM and RDM than in both pre-treatment. Conclusion: The increased activity and expression of 11β-HSD1 visceral bed fat may be one of the mechanism of T2DM and metabolism syndrome.
