Quantitative Analysis of Collagen Components in Mesangial Areas of Mesangial Proliferative Glomerulonephritis in Adults

Objective:To study the expressions and quantitative change of collagen components, such as collagen type Ⅰ and collegen type Ⅳ . in Mesangial areas of mesangial Proliferative Nephritis (MsPGN) in adults. Methods:We applied immuno-histochemical staining assay (streptavidin-peroxidase, SP) to study the expressions of collagen type Ⅰ and collagen type Ⅳ, used image analysis system (HPIAS-1000) to analysis the stained surface areas of the two collagen components. Results: The two component expression difference between grave MsPGN and mild or media MsPGN have significance (P 0. 05), and the difference between expressions of collagen type Ⅰ or collagen type Ⅳ in mild and media proliferative mesangial areas also have significance (P0. 05). The expressions of collagen type Ⅰ and collagen type Ⅳ have positive correlation, whose coefficient was 0.628. Conclusion: This study have demonstrated that the expressions of collagen type Ⅰ and collagen type Ⅳ increase in mesangial areas with the pathological changes getting intenser, the two collagen expression tensity is different in grave .media or mild proliferative mesangial areas. Therefore, the collagen component analysis can be used as one of the standards that can indicate the degree of pathological change in MsPGN.
