COMPARINSON BETWEEN FPIA AND RIA FOR MOINITORING THE CONCENTRATION OF CYCLOSPORINE A IN BLOOD

A comparison between Fluorescence Polarization Immunoassay (FPIA) and Radioimmunoassay(RIA) is reported for Cyclosporine A blood concentration monitoring. The results showed that both correlated well in whole blood or serum (RIA = 0.773 FPIA + 66. 8,r= 0. 94;RIA = 0. 378 FPIA + 17. 00,r=0. 96), with P less than 0. 05(P0. 05). The mean FPIA/RIA ratio (X±SD) was 1. 05± 0. 26 in whole blood,and 2. 42 ± 0. 73 in serum.Due to lack of speciality, FPIA, fast automated method as it is, fails to differentiate individual components among a mixture of metabloites of Cyclosporine A in blood in monitoring practice, it would be better to combine with other methods such as RIA,HPLC etc.
