A buffered aspirin tablet was designed and its in vitro dissolution rate, in vitro acid-neutralizing capacity single dose and multiple dose bioavail-abilities and irritation on gastric mucosa were studied.The rate of aspirin dissolution was significantly increased with buffered tablet. It resulted slightly higher peak concentration, slightly larger AUC and significantly earlier peak time compared to that with common tablet, following oral administration in the fasting state. Cm were 154 and 133 μg/ml, AUC were 1 454 and 1 343 μg/ml · h, Tm were 1. 23 and 3. 08 h respectively. The maximum salicylate concentration at steady-state after multiple dose of buffered aspirin tablet and common tablet were 297. 5 and 270μg/ml respectively. The examination of the stomach mucosa was carried under a dissecting microscope 2 h after administration of drug to rats. It was found that the acute gastric erosion following administration of aspirin buffered tablet was significantly reduced compared with common tablet.It is concluded that buffered aspirin tablet causes less gastric irritation, dissolves more rapidly, and is absorbed much better without decreasing the steady-state concentration.
