Screening of high-yield strain for amylase by induced mutation

Bacillus strain as an original strain screened from Jiande of Zhejiang was mutated by UV,diethyl sulfate and the two alternate treatments to select species for high amylase production.Starch iodine color method was used for the primary screening,that is,H/C(ratio between diameter of clear zone and diameter of strain circle) greater strains were picked from starch medium plate.Then they were re-screened by shaking flask and activity of amylase in fermentation fluid.The results indicated that the activity of amylase produced by mutated strain B2 with genetic stability was 2.01 U/mL,which was 1.31 times as high as that produced by the original strain.
