Process optimization and model for decalcification of bighead carp fish bone

The pretreatment study of decalcification of bighead carp fish bone was carried out in order to fluently extracting purer collagen from it. Single experimental method and response surface experimental method were used to investigate the effects of main process parameters of ratio of liquid to solid,hydrochloric acid mass fraction,stirring speed and stirring time on fish bone decalcification rate. The parameters were optimized to be liquid/solid ratio7. 3∶1 g·g-1,hydrochloric acid mass fraction 7. 0 %,stirring speed 1 000 r·min-1 and stirring time 1. 0 h,respectively,under which the decalcification rate reached up to 91. 78%. The model with the four parameters as independent variables and decalcification rate as response value was established,whose theoretical values have good agreement with experimental values and can be used to predict decalcification rate at different process conditions. The significant degree of effects of process parameters on decalcification rate were determined and compared,which can provide the theoretical basis for strictly controlling the operation indexes.
