Effect of heparin on myocardial mitochondria ultrastructure following asphyxia cardiac arrest-resuscitation in rats

Objective To investigate the effects of heparin on myocardial mitochondria ultrastructure injury induced by asphyxial cardiac arrest-resuscitation in rats. Methods Sixty-nine SD rats weighing 450 ~ 500 g were randomly divided into 3 groups: the sham group(n = 6),the control group(n = 30),the heparin group(n = 30). The control and heparin groups were subdivided into0. 5-hour,2-hour,and 6-hour subgroups,each group had 10 rats. The asphyxial cardiac arrest model was prepared. The heparin group was received conventional resuscitation while intravenous heparin 0. 5 mg/kg. The control group was intravenously injected with the same volume of saline,and the sham group was only operated with intravenous and tracheal intubation. After 0. 5 hours,2 hours and6 hours of the restoration of spontaneous circulation,the specimens were prepared for electron microscopy and semi-quantitatively analyzed by the Flameng method. Result There was no significant difference in body weight(W),rectal temperature(T),mean arterial pressure(MAP),heart rate(HR),and end-tidal carbon dioxide(PETCO2) before resuscitation in the rats. The Flameng score of the mitochondria increased at 0. 5,2 and 6 hours after resuscitation in Heparin and the worst was at 2 hours after resuscitation. However,the mitochondrial semiquantification of Flameng after the resuscitation decreased in the heparin group(P 0. 05). Conclusion Cardiac arrest-cardiopulmonary resuscitation causes the myocardial mitochondrial damage. Heparin can reduce myocardial mitochondrial damage.
