The Preparation and preliminary evaluation of quality of matrine sustained-release pellets

Objective To prepare matrine sustained-release pellets by extrusion-spheronization and fluid bed coating technology and investigate the property of pellets by micromeritic evaluation parameters and studying its in vitro releasing. Methods The pellet core with microcrystalline cellulose(MCC) were prepared by the process of extrusion-spheronization. The Ballability pellets(surface morphology,roundness,bulk density and yield) was looked as index,prescription and adhesive of pellets were screened. The evaluation of the pellets in vitro release was evaluated. Results The prescription of sustained-release pellets was set as follows: 25%weight gains of matrine,5% chitosan,62% MCC,8% octadecanol. The optimum technologic parameters of pellets were as follows: the rate of extrusion was 43/min and the time of spheronization was 10 min. The enteric coating technologies were as follows: the material temperature at 40 ℃,the inlet wind temperature at 58℃ and the atomization pressure 0. 18 MPa. Through this process the matrine pellets drug loading rate was 90%,7 h cumulative release rate in artificial colonic fluid was 94. 23%. Conclusion Matrine sustained-release pellets is easily prepared,economical and is characterized by delayed-release.
