Determination of Voriconazole in the Human plasma by HPLC for Clinical application

Objective To establish the determination method of voriconazole the in Human Plasma by HPLC,and apply it to individualized treatment.Methods Blood samples were pretreated by using protein precipitation.Analytical column was Gemini C18(4.6 mm × 150 mm,5 μm),mobile phase was acetonitrile-glacial acetic acid(40∶60) at a flow rate of 1.0ml/min.Detection wavelength was set at 255 nm and temperature was kept at 35 ℃.Results Voriconazole showed good linear relationships in ranges of 0.227~ 13.540 μg/ml(r=0.999).Absolute recoveries of voriconazole were above 102.8%~113.6%,and relative recoveries were 106.3%~111.4%.Intra-day and inter-day RSD values were less than 5%.Conclusion The HPLC method is simple,quick and sensitive for drug monitoring on voriconazole.High concentration variability was observed among patients,and routine monitoring of voriconazole blood level may improve the drug safety.
