Autophagy is upregulated under acidic stress in gastric cancer cell line SGC7901

Objective To investigate the effects of acidic stress on the autophagy of SGC7901 cells.Methods The human gastric cancer cell line SGC7901 was selected as the cell to be studied and the gastric cancer SGC7901 cells were treated in complete medium,adjusted pH to 6.5 or pH 7.4.Immunofluorescence was used to detect the formation of autophagosome in SGC7901 cells under acidic stress.The expression of LC3 was detected by Western Blot.Results The observation under Laser confocal electron microscopy demonstrated that the number of fluorescent granules in SGC7901 cells treated with acidic stress was significantly higher than that in the control group.The expression of LC3-Ⅱ in cells treated with acidic medium was higher than that in control group and a decreased in expression of p62 in SGC7901 cells under acidic medium environment was observed.Conclusion Acid stress can cause autophagy enhancement in gastric cancer SGC7901 cells.
