Activation and significance of TGFβ1/smad7 signaling pathway in rat's hepatic fibrosis

Objective To investigate the activation and significance of TGFβ1 / smad7 signaling pathway in hepatic fibrosis of rats. Methods Wistar rats(n =40) were divided into control group and model group(each group,n=20). The rats were injected with CCl4 to induce liver fibrosis. Serum TGFβ1,ALT and HA were detected by enzymic method and radio-immunity. Liver inflammation and liver fibrosis was graded by HE staining. The expression of TGFβ1,Smad3,Smad7 of liver tissue were examined by Western blot. The expression of TGF-β1,Smad3,Smad7 m RNA were analyzed by RT-PCR. Results Compared with control group,the expression of TGFβ1(3.87±0.87),Smad3(2.64±0.95),proteins and TGFβ1(0.87±0.05),Smad3(0.53±0.04) m RNA were increased but Smad7(1.21±0.54) protein and Smad7(0.34±0.03) m RNA were decreased in model group. The level of TGFβ1,ALT and HA was higher in model group than that of the control group. Conclusion TGFβ1 / smad7 pathway overactivition and increase of serum TGFβ1,ALT and HA may closely be related to liver fibrosis formation development.
