Feasibility of the dried blood spots specimen method used to screen prenatal Down's syndrome among mid-term pregnant women

Objective To explore the feasibility of the dried blood spots specimen methods used to screen prenatal Down' s syndrome among mid-term pregnant women. Methods The levels of serum Alpha-fetoprotein(AFP) and free beta-human chorionic gonadotropin(β-h CG) in dried blood specimen on filter paper were detected with the method of CLEIA. The variation coefficient, recovery rate and detection limit were estimated.The screening result of serum AFP and β-h CG in dried blood specimen on filter paper specimen methods was compared with that of routine serum methods. Results In test AFP,the variation coefficient,recovery rate and detection limit was 2.1%-6.5%,96.2% and 0.5 ng / ml,respectively;in test β-h CG,the corresponding results was3.9%-6.7%,98.4% and 0.5 ng / ml,respectively. The onset risk rate was 1 / 128,with an accordance rate of 98.89%compared with that of the gold standard of serum screening method which is universally accepted. A total of1260 pregnant women were screned for prenatal Down's syndrome 74 were positive(5.9%) among whom one fetus was diagnosed as Down's syndrome by detecting the chromosomes in the amniotic fluid. Conclusion The method of CLEIA through dried blood spots specimen,substitutes for the routine serum method to detect the levels of AFP and β-h CG among mid-term pregnant women is convenient and feasible,and its screening result is accurate and reliable.
