Analysis of correlation between p28GANK protein expression and liver cancer pathogenesis

Objective To analyze the correlation between the expression of p28 GANK protein and the pathogenesis of primary liver cancer(PLC) and its possible role mechanism. Methods The paraffin embedded tissue specimens of 48 patients with PLC who underwent surgical resection and confirmed by pathology in Henan Provincial Infectious hospital from March 2015 to March 2018 were collected. Among them,28 patients were sampled adjacent tissues during surgery,and all of them were fixed with 10% formal dehyde solution,paraffin embedded and sliced. The p28 GANK protein expression level was analyzed by immunohistochemistry SP staining method. At the same method,the expression levels of apoptosis related-genes Bcl-2 and Bax proteins in tissue samples were evaluated and given statistical analysis. Results The positive expression of p28 GANK protein was mostly brown and yellow granules,located in the nucleus and cytoplasm. The positive expression rates of p28 GANK protein,Bcl-2 and Bax proteins in PLC tissues were significantly higher than those in adjacent tissues(P0.05). Spearman level correlation analysis shows there was a significant correlation between the expression of p28 GANK protein and the positive expression of Bcl-2 protein in PLC tissues(P 0.05),but there was no correlation with the positive expression of Bax protein(P0.05). There was no significant difference in the positive expression rate of p28 GANK protein in PLC patients with different gender,age,maximum tumor diameter and liver function grading(P0.05),but the positive expression rate of p28 GANK protein in patients with stage Ⅲ-Ⅳ was significantly higher than that of patients with stage Ⅰ-Ⅱ,and the positive expression rate of p28 GANK protein in patients with lymph node or distant metastasis was obviously higher than that in patients without lymph node or distant metastasis(P 0.05). Conclusion The expression of p28 GANK protein is related to the occurrence and development of PLC:the high expression of p28 GANK protein may affect the invasion and metastasis of tumor by up-regulating the expression of apoptosis inhibitory gene Bcl-2.
