Growth inhibition effects of D-limonene on Human Gastric Carcinoma MGC803 cells

Objective To investigate the antieancer effect of D-limonene on Human Gastric Carcinoma MGC803 cells.Methods Cell growth inhibition was measured by MTT colorimetric assay;apoptosis rate was measured by flowcytometry(FCM);laser scanning confocal microscopy (LSCM)was used to detect the reactive oxygen species(ROS)and the expression status of caspase-3 proteins.Results Growth inhibition of D- limonene on MGC803 cells was in a dose dependent manner;FCM detected that d-limonene have induced apoptosis in a time depedent fashion;The generation of reactive oxygen speeies(ROS)as well as the expression of caspase-3 proteins which were examined by LSCM,they were both increased obviously(P0.05)after D-limonene treated the cells.Conclusion D-limonene significantly inhibited the proliferation and induced apoptosis of MGC803 cells.lt is likely that D-limonene associated with the gerneration of ROS within cells,and changing the expression of caspase-3 level may be one of the anfieancer mechanisms.
