Effects of Recombinant Human Erythropoietin on The Expressipn of The Proliferating Cell Nuclear Antigen in Endothelial Cells from Human Umbilical Veins

Objective To study the effects of recombinant human erythropoietin on the the proliferation of endothelial cells from human umbilical veins.Methods The endothelial cells were isolated from human umbilical vein and cultured with 10U/ml,20U/ml and 40U/ml rHuEPO,respectively.Then the expression of proliferating cell nuclear antigen(PCNA) was detected using immunohistochemical methods.ResultsThe positive percentage of PCNA in HUVECs from control group,10U/ml rHuEPO group,20U/ml rHuEPO group and 40U/ml rHuEPO group were(31±3)%,(40±5)%,(68±6)% and(65±5)%,respectively.Compared with the control group,the proliferation of endothelial cell in rHuEPO group enhanced significantly(P0.05)in a dose-dependent manner.Conclusion rHuEPO can promote the proliferation of endothelial cell cultured in vitro.
